NextGen-HLA typing is a Next Generation Sequencing (NGS) based high-resolution identification of Human Leucocyte Antigen (HLA). The HLA system is a complex of genes on chromosome 6, which encode cell-surface proteins responsible for regulation of the immune system also known as Major histocompatibility complex (MHC) proteins. There are two distinct classes. Class I (A, B, C) & Class II (DR, DQ & DP). HLA genes are highly polymorphic and play an important role in immune – mediated diseases, tumour development processes, transplanted organ or tissue survival determination and drug hypersensitivity. They are particularly important in the case of transplanted tissue, because it can lead to transplant rejection & hence the HLA typing is done to determine HLA match between the Donor & the Recipient. NGS offers Higher & better match between donor & recipient influencing successful outcomes for Hematopoietic stem cell (HSCT) & any solid organ transplant. NGS helps in removing all ambiguity & provides most accurate characterisation of alleles. All the 6 loci shall be reported with resolution up to 6 digits.

Frequently Asked Questions
Human Leukocyte Antigens (HLA) typing Ensures the growth and development of transplanted new healthy cells. Reduces the risk of a post-transplant complication of graft-versus-host (GVHD) disease. To find the best match for donor selection in allogeneic bone marrow , solid organ, tissue transplant.
Solid Organ Transplantation requires antigen level typing for HLA-A, B, and DR, DP , DQ Antigens.
HLA –Human Leukocyte antigen are cell surface antigen that induce an immune response, recognize and eliminate foreign cells/ antigens that enter the body. They are the primary source of immune-rejection during an organ transplant leading to rejection of allograft. Every individual has a distinct HLA pattern inherited from their parents. Offspring's will be HLA haplo-identical inheriting one pattern from each of their parents. Identical HLA typing - complete matching of all alleles in both donor and recipient‘s
